ABSTRACT

Background:Phenol-sulfuric acid assay has been widely used for quantification of sugars in biological fluid and industry. The conventional method originally proposed by Dubois et al, was modified several times for enhancing sensitivity of assay such as substituting phenol with other chromogens, by optimizing assay conditions and by adding phenol after dehydration reaction. Both conventional and modified assays have utilized acid catalysis property of sulphuric acid but effect of adding phosphoric acid has not been studied before. Methods:The present study was conducted in Department of Biochemistry, ANIIMS, Port Blair and IGIMS, Patna. The method being developed in our study consisted of adding orthophosphoricacid to the reaction mixture before addition of sulphuric acid. The method was optimized with different amounts of orthophosphoric acid. Statistical analysis was done using SPSS and Microsoft Excel software.Results:The current study found an enhancingeffect (on sensitivity) of orthophosphoric acid in optimal concentration of 5.16 mmoles. Comparison among standard curves of methods that were compared showed that the curve was steepest for current study and average absorbance was 0.199±0.017 for conventional, 0.253±0.011 for method by Rasouli et al,and 0.290±0.013 for current study. Pooled serum analysis exhibited absorbance of 0.157±0.015 in conventional method while in modified conventional method absorbance was 0.234±0.010 and highest absorbance was observed in current study at 0.281±0.012.Conclusions:Our results suggest that orthophosphoric acid exerts a positively modifying effect on phenol sulphuric acid assay.

ABSTRACT

Objective: To establish a method for determining the content of total polysaccharides in decoction pieces of Polyporus,analyze the content of total polysaccharides in samples with different sources and grades. Method: The relative molecular weight and the polydispersity index of polysaccharides in decoction pieces of Polyporus were measured by a high performance gel chromatography coupled with a multi-angle laser light scattering and refractive index system.Dextran with similar molecular weight as polysaccharides was selected as the reference substance.Orthogonal experiment and single factor tests were used to optimize the pretreatment conditions for the determination of total polysaccharides in Polyporus.Polysaccharides in Polyporus with different areas and grades were determined by anthrone-sulfuric acid colorimetric method at 630 nm. Result: The linearity,stability,precision,repeatability and recovery rate of the established method all reached the standards,respectively.The content of total polysaccharides in samples from different areas ranged from 0.87% to 1.39%.The content of total polysaccharides in samples with different grades was 1.40% for first-grade pieces,1.21% for second-grade pieces, and 1.03% for third-grade pieces. Conclusion: The established method is simple,accurate and reproducible,and it can be used for the determination of polysaccharides in decoction pieces of Polyporus.The content of polysaccharides in samples from different origins varies greatly.The content of polysaccharides in samples with different grades shows a certain regularity.The content of polysaccharides is the highest in the first-grade pieces,followed by the content in the second-grade,and the lowest in the third-grade.The results can provide a reference for formulating limits for the content of total polysaccharides and the grade standard of decoction pieces of Polyporus.

ABSTRACT

Objective :To optimize the extraction of polysaccharide from the residue of Periplaneta americana by response surface method and study the effect of P. americana polysaccharide (PAP) on wound healing, so as to provide ideas for the comprehensive utilization of P. americana residues. Methods On the basis of single factor experiment, Box-Behnken response surface method was used to design the best extraction process. And the monosaccharide composition of polysaccharide was analyzed. Polysaccharide concentrations of 0.1, 0.05, and 0.025 g/mL were applied to the wound injury model mice. The wound healing rate was calculated, and hematoxylin-eosin (HE) and Sirius Red staining were used to evaluate the wound healing degree. Results :The optimum extraction technology of PAP were as follows: The concentration of NaOH was 0.02 mol/L; And the extraction time was 1.65 h; The extraction temperature was 62 ℃. The transfer rate of polysaccharide extracted by this process was 13.68%. The polysaccharide of the P. americana consisted of seven monosaccharides of mannose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, and fucose. Wound healing experiment showed that wound healing rate of the polysaccharide group on day 3, 6, 9, and 14 was higher than that of the model group, and HE and Sirius Red staining showed that the polysaccharide could promote the formation of collagen and granulation tissue, thus promoting the wound healing. Conclusion: The optimized extraction process of PAP from P. americana residues by response surface method was reliable, with high extraction rate. And PAP could promote wound healing in mice, suggesting that the polysaccharide from P. americana residues could be used as wound related products.

ABSTRACT

OBJECTIVE: To establish a determination method for content of polysaccharide in Acanthopanax senticosus, and to conduct cluster analysis and ultrasonic extraction technology optimization. METHODS: The content of polysaccharide in A. senticosus was determined by phenol-sulfuric acid method. Cluster analysis was conducted by using SPSS 23.0 software of the polysaccharide in A. senticosus from 17 habitats. The ultrasonic extraction technology was optimized with L9(34)orthogonal test using extraction temperature, the ratio of material to liquid, extraction time as factors, the content of polysaccharides as index, and then validated. RESULTS: The linear range of glucose ranged from 0.007 75 to 0.151 mg/mL (r=0.999 1); the limits of quantification and detection were 2.854, 0.856 μg/mL; RSDs of precision, stability, and repeatability tests were less than 2%; recovery rates of the sample were 98.41%-101.58%(RSD=1.23%,n=6). Cluster analysis results showed that 17 batches of samples could be clustered into three classes; S1, S6, S10, S12 and S13 were clustered into one class; S3-S5 and S7 were clustered into one class; and the rest samples were clustered into one class. The optimal ultrasonic extraction technology was extraction temperature 55 ℃, ratio of material to liquid 1 ∶ 10 (g/mL), extraction time 35 min. Results of validation tests showed the content of the polysaccharide under the best process condition was 4.36%(RSD=0.920%,n=3). CONCLUSIONS: Established method is simple,reproducible and suitable for determination of polysaccharide in A. senticosus; the optimized ultrasonic extraction technology is stable and feasible.

ABSTRACT

SATB1 plays a crucial role in the invasion and metastasis of breast cancer,and inhibition of SATB1 expression can effectively control breast cancer metastasis. In this study,homogeneous polysaccharides were isolated from Poria cocos and their sulfated derivatives were prepared to screen out the polysaccharide compositions with inhibitory effects on SATB1 expression. Smal-molecule components were removed from P. cocos by ethanol extraction,and P. cocos crude polysaccharide PPS was obtained by water extraction and ethanol precipitation. Then PPS was successively separated by DEAE Sepharose fast flow anion-exchange and Superdex-75 gel permeation chromatographic steps to give PPSW-1. The structure of PPSW-1 was identified and its sulfated derivatives were prepared. Then their inhibitory effects on human breast cancer MDA-MB-231 cells were investigated. A kind of polysaccharide,PPSW-1 with inhibitory effect on human breast cancer MDA-MB-231 cells,was obtained from P. cocos,with a relative molecular weight of 3. 06×104,and structure of 1,6-branched 1,3-α-D-galactan. PPSW-1 and its sulfated derivative Sul-W-1 showed good inhibitory effect on cells migration,and the water solubility of Sul-W-1 was better than that of PPSW-1. In addition,it was found that polysaccharide of P. cocos and its sulfated derivative can inhibit expression of SATB1. In this study,a kind of homogeneous polysaccharide with inhibitory effect on human breast cancer MDA-MB-231 cells was isolated from P. cocos,and its sulfated derivative with similar efficacy but better solubility was prepared,laying the foundation for the substance basis study of P. cocos.

ABSTRACT

ABSTRACT Portable X-ray fluorescence spectrometer (pXRF) has been recently adopted by the Soil Science community for uses in both field and laboratory, obtaining the total content of several chemical elements in a few seconds. Sulfuric acid digestion is an expensive and time-consuming laboratory analysis that provides contents of Fe2O3, Al2O3, SiO2, TiO2 and P2O5, important for soil studies. Due to few pXRF studies in tropical soils, this work aimed to compare contents of Fe2O3, Al2O3, SiO2, TiO2 and P2O5 obtained by pXRF with sulfuric acid digestion results, and to evaluate the effects of varying forms of preparing soil samples and scanning with pXRF on the resulting values in Brazilian soils. Soils were scanned in five conditions in-field (in situ) and in laboratory, evaluating varying sample preparation methods, particle sizes and soil moisture. Four pXRF scanning operational modes were tested. Linear regressions were adjusted between results of pXRF and sulfuric acid digestion. Equations were validated with an independent set of samples. Statistical analyses compared the methods of preparing the samples. Adequate linear models reached R2 of 0.99 and 0.89 for Fe2O3 and TiO2, respectively. Validation promoted R2 greater than 0.97 and RMSE and ME close to zero for both oxides. Statistical differences of pXRF results were found among the methods of preparing samples. pXRF spectrometer has great potential to obtain Fe2O3 and TiO2 content rapidly and economically with high correspondence with laboratory results of sulfuric acid digestion analysis. Varying methods of preparing the samples promote differences in the results of pXRF.


RESUMO O espectrômetro portátil de fluorescência de raios-X (pXRF) foi recentemente adotado pela Ciência do Solo, para uso em campo e laboratório, para obtenção do conteúdo total de vários elementos químicos em poucos segundos. A digestão com ácido sulfúrico é uma análise laboratorial cara e demorada que fornece teores de Fe2O3, Al2O3, SiO2, TiO2 e P2O5, importantes para estudos sobre solos. Devido aos poucos estudos sobre o pXRF em solos tropicais, este trabalho objetivou comparar os teores de Fe2O3, Al2O3, SiO2, TiO2 e P2O5 obtidos pelo pXRF com os resultados de digestão com ácido sulfúrico e avaliar os efeitos de diferentes formas de preparo de amostras de solo e leitura com o pXRF sobre seus resultados para solos brasileiros. Os solos foram submetidos a leituras com o pXRF em cinco condições, em campo (in situ) e em laboratório, avaliando variados métodos de preparo de amostras, tamanhos de partículas e umidade do solo. Quatro modos de operação do pXRF foram testados. Regressões lineares foram ajustadas entre os resultados do pXRF e digestão com ácido sulfúrico. As equações foram validadas com um conjunto independente de amostras. Análises estatísticas compararam os métodos de leitura de amostras. Modelos lineares adequados atingiram R2 de 0,99 e 0,89 para Fe2O3 e TiO2, respectivamente. A validação promoveu R2 maior que 0.97 e RMSE e ME próximos a zero para ambos os óxidos. Foram encontradas diferenças estatísticas dos resultados do pXRF entre os métodos de preparo de amostras. O pXRF possui um grande potencial para obter rápida e economicamente os teores de Fe2O3 e TiO2 com elevada correspondência com os resultados laboratoriais da análise da digestão com ácido sulfúrico. Métodos variáveis ​​de preparo das amostras promovem diferenças nos resultados de pXRF.

We reviewed the chemical/physical properties, toxicity, environmental fate, and ecotoxicity of strong acids in soil and groundwater environments. We recommend that sulfuric acid and hydrofluoric acid be classified as chemicals of priority control based on volumes used, toxicity, carcinogenicity, and past significant spill events. Understanding the behavior and transport of spilled strong acids in soil and groundwater environments requires a multi-disciplinary approach, as they can undergo a variety of geochemical and biochemical reactions with complex geomedia. The toxicity of spilled acid is dependent on the characteristics of the geomedia exposed to the acid and the amount of residual protons following acid–substrate interaction. Soil texture, cation exchange capacity, mineral composition, bedrock type, and aluminum content may be important factors affecting the toxicity of spilled acid in soil-groundwater environments. We expect that the results of this study will contribute preliminary data for future research on chemical spills.

ABSTRACT

The seaweeds are bio-resource rich in sulfated and neutral polysaccharides. The tropical seaweed species used in this study (Solieria filiformis), after dried, shows 65.8% (w/w) carbohydrate, 9.6% (w/w) protein, 1.7% (w/w) lipid, 7.0% (w/w) moisture and 15.9% (w/w) ash. The dried seaweed was easily hydrolyzed under mild conditions (0.5 M sulfuric acid, 20 min.), generating fermentable monosaccharides with a maximum hydrolysis efficiency of 63.21%. Galactose and glucose present in the hydrolyzed were simultaneously fermented by Saccharomyces cerevisiae when the yeast was acclimated to galactose and cultivated in broth containing only galactose. The kinetic parameters of the fermentation of the seaweed hydrolyzed were Y(P/S) = 0.48 ± 0.02 g.g−1, PP = 0.27 ± 0.04 g.L−1.h−1, η = 94.1%, representing a 41% increase in bioethanol productivity. Therefore, S. filiformis was a promising renewable resource of polysaccharides easily hydrolyzed, generating a broth rich in fermentable monosaccharides for ethanol production.


As algas marinhas são recursos naturais ricos em polissacarídeos sulfatados e neutros. A espécie de macroalga tropical utilizada neste estudo (Solieria filiformis) apresentou teores de carboidratos de 65,8% (m/m), proteínas de 9,6% (m/m), lipídios de 1,7% (m/m), umidade de 7,0% (m/m) e 15,9 % (m/m) de cinzas. A macroalga seca foi facilmente hidrolisada em condições brandas, na presença de ácido sulfúrico 0,5 M, por 20 min, produzindo monossacarídeos fermentáveis com uma eficiência de hidrólise máxima de 63,21%. A galactose e a glicose presentes no hidrolisado foram fermentadas simultaneamente por Saccharomyces cerevisiae, após aclimatação da levedura cultivada em meio contendo apenas galactose como fonte de carbono. Os parâmetros cinéticos da fermentação do hidrolisado algáceo pela levedura aclimatada a galactose foram Y(P/S) = 0,48 ± 0,02 g.g-1, PP = 0,27 ± 0,04 g.L- 1.h-1, η = 94,1%. Portanto, a macroalga S. filiformis se mostrou um recurso renovável promissor como fonte de polissacarídeos facilmente hidrolisados, gerando um meio nutritivo rico em glucose e galactose para a produção de etanol.

ABSTRACT

OBJECTIVE:To establish a method for the content determination of saccharide in polysaccharides containing galact-uronic acid based on phenol-sulfuric acid method combined with correction factor method. METHODS:The determination condi-tion of phenol-sulfuric acid was optimized. A series of standard curves were drawn with glacturonic aid-glucose mixed control with different mass ratio. The content of saccharide in Codonopsis pilosula polysaccharides CPP1b samples containing galacturonic acid was determined. According to regression equation of galacturonic acid-glucose ratio of 0-100%,the correction factor was calculated by using C. pilosula polysaccharides CPP1b as the reference polysaccharide,and the results of content determination of saccharide were corrected. The rationality of this method was verified by using mixed monosaccharide control with same composition as C. pi-losula polysaccharides CPP1b. RESULTS:The correction factor of C. pilosula polysaccharide CPP1b to glucose was 3.33;in vali-dation test,the content of saccharide in mixed monosaccharide control with same composition as C. pilosula polysaccharides CPP1b was 103.47%. RSDs of precision and stability tests was <1%. The recoveries ranged 93.52%-107.35%(RSD=5.09%,n=6). CONCLUSIONS:The established method can accurately determine the content of saccharide in C. pilosula polysaccharides CPP1b containing galacturonic acid.

ABSTRACT

Objective To establish the assay method for the total polysaccharide in Pudi Enema.Methods Phenol-sulfuric acid method was used for chromogenic reaction.The content of total polysaccharide was measured by UV spectrophotometry at 488.8 nm.Results The total polysaccharides calibration curve was at the range of 0~22.635 mg/L, with regression function being Y=0.062 06 X-0.003 34(r=0.999 8).The recovery of calycosin was 98.36%(RSD=2.34%).Conclusion This method is sensitive,rapid,accurate and reliable.It can be used to assay the content of total polysaccharide in Pudi Enema.

ABSTRACT

Design space approach was applied in this study to optimize the lime milk precipitation process of Lonicera Japonica (Jinyinhua) aqueous extract. The evaluation indices for this process were total organic acid purity and amounts of 6 organic acids obtained from per unit mass of medicinal materials. Four critical process parameters (CPPs) including drop speed of lime milk, pH value after adding lime milk, settling time and settling temperature were identified by using the weighted standardized partial regression coefficient method. Quantitative models between process evaluation indices and CPPs were established by a stepwise regression analysis. A design space was calculated by a Monte-Carlo simulation method, and then verified. The verification test results showed that the operation within the design space can guarantee the stability of the lime milk precipitation process. The recommended normal operation space is as follows: drop speed of lime milk of 1.00-1.25 mL•min⁻¹, pH value of 11.5-11.7, settling time of 1.0-1.2 h, and settling temperature of 10-20 ℃..

ABSTRACT

Objective:To study the boiling water brewing extraction of astragalus polysaccharides in order to solve the problems of long cycle , high energy consumption and poor clarity in the traditional extraction , and lay foundation for the industrial production . Methods:Using the extraction yield of astragalus polysaccharide as the evaluation index and phenol -sulfuric acid method applied to de-termine the content of astragalus polysaccharide , the boiling water brewing extraction was used for astragalus polysaccharides .The effects of brewing time , boiling water amount and brewing times on the polysaccharide content were studied by single factor experi -ments, and then an orthogonal design method was used to screen the optimum technology parameters .The traditional water decoction extraction process was employed as the control .Results:The optimum conditions of boiling water brewing were as follows:adding 9-fold amount of water and soaking 60 min for three times.The astragalus polysaccharides extraction yield (4.81) of the optimal technology was higher than that of the traditional water decoction extraction (4.06%).Conclusion:Boiling water brewing method used to extract astragalus is with high extraction yield , simple operation , short cycle and low energy consumption , the color of astragalus polysaccha-ride is light, and it is clear after dissovled in water , which is superior to the traditional water decoction extraction method , and provide a new method with broad application prospect for the preparation of astragalus related preparations .

Objective To optimize the technological conditions of polysaccharide purification from compound Qianyu water decoction. Methods Water extraction and alcohol precipitation, resolution, Sevag method and dialysis method were used to purify polysaccharide.The purity of polysaccharide was measured with the phenol-sulfuric acid spectrophotometry.On the basis of single factor test, effects of redissolved solid-liquid ratio, number of protein removal, and dialytic time on polysaccharide purity of Qianyu were investigated by orthogonal test. Results The best conditions for purification of polysaccharide in Qianyu were as follows: liquid-solid ratio was 1:40(g/mL, W/V), remove protein for 10 times, and dialysis for 18 h.The content of polysaccharide could reach 69.04%, and the transfer rate was 51.84%. Conclusion The optimized purification process was simple and accurate.It can be used for polysaccharide purification in compound Qianyu water decoction.

ABSTRACT

PURPOSE: This study was conducted to identify the characteristics associated with sulfuric acid injury in the emergency department. METHODS: Data were collected retrospectively from January 2007 to December 2015 on all sulfuric acid injuries presenting to the emergency department in Gu-mi Soonchunhyung University Hospital. Patients injured by sulfuric acid were recorded over a nine year study period and collected data included demographics, injury mechanism, injured body part, hospital care and final diagnosis. RESULTS: A total of 26 cases were identified. Most patients were male (88.5%) and the face was the most commonly injured body part. The most common mechanism of injury was splashing injury. A total of 16 (61.5%) patients were identified as having lesions worse than second degree burns. CONCLUSION: Sulfuric acid can cause severe and fatal skin burn. When working with sulfuric acid, acid proof protect clothing, goggles and glove should be worn. Furthermore, safety education and workplace environment improvement are necessary to reduce sulfuric acid injury.

ABSTRACT

Objective: Panax notoginseng is an important Chinese medicinal plant. Saponin accumulation is higher in the flower buds than in the other parts of P. notoginseng. However, the flower bud compositions have not yet been quantified. The aim of this study is to investigate the formation and accumulation of saponins in the flower buds of P. notoginseng from different populations and at different growth years. Methods: Fourteen types of P. notoginseng with different growing durations and from different areas of Wenshan County, Yunnan Province were collected. We separated P. notoginseng individually into the flower buds, stems, leaves, and roots at the places where it has the highest saponin content. An efficient high-performance liquid chromatography (HPLC) method was developed for simultaneously quantifying two active saponins, ginsenoside Rb1 and ginsenoside Rb3, in the flower buds of P. notoginseng. The total saponin content was determined by using a quantitative vanillin-sulfuric acid colorimetric method. HPLC method was used to establish the fingerprints of 13 saponins and then quantify the composition in the whole plant of P. notoginseng. Results: The saponin contents of different parts differ significantly, and the total saponin content and those of Rb1 and Rb3 do not entirely correlated. The flower buds of P. notoginseng contain 27.79% of total saponins, which is the highest saponin content in the whole plant. Fingerprint result showed that different saponins were appeared in different parts of the plant, i.e. flower buds, stems, leaves, and roots. Conclusion: The saponin contents from the flower buds of P. notoginseng vary depending on the growth area and duration. The fingerprints show that the saponin contents and compositions vary depending on the part of P. notoginseng. These results are useful for the pharmacological evaluation and quality control of P. notoginseng.

ABSTRACT

A poly( GMA-EGDMA) coated SPME fiber was prepared using an in-situ polymerization by direct bonding to the surface of a polydopamine-modified stainless steel wire. Then the fiber was modified by sulfuric acid. A novel solid phase microextraction coating coupled to high performance liquid chromatography ( HPLC) method based on the as-prepared fiber was developed for the determination of four pharmaceuticals and personal care products ( PPCPs) in water samples. The influences of extraction parameters, including pH, extraction time, extraction temperature and salt addition were investigated. 3 mL water sample was extracted by the as-prepared fiber for 60 min at 30 ℃, and then desorbed with mobile phase for 30 min, respectively. Desorption solution was analyzed by HPLC-DAD ( diode array detection ) . The results indicated that the extraction yield of the fiber was good for four PPCPs. The linear correlation coefficients were>0. 997 with the linear range of 2-200 μg/L. The limits of detection (S/N=3) were 0. 5-5 μg/L with RSD (n=5) of 4. 1%-11. 9%. The recoveries of four PPCPs at spiked level of 20, 50, 100 μg/L were within the range of 70. 6%-105. 5%. The results showed that this method was easy, green, accurate and precise, and could be used to assay the four PPCPs in real water samples.

ABSTRACT

By studying on extraction technology of desertliving cistanche polysaccharides, it provided evidences for development and utilization of cistanche. Anthrone-sulfuric acid method was used in the measurement of desertliving cistanche polysaccharides. The yield of desertliving cistanche polysaccharides was used as index. Different types of extraction methods were compared. Then, the extraction process was optimized by orthogonal experiment. The results showed that the best extraction process was Tween-60 ultrasonic cooperated. The best extraction process was when the mass concentration of Tween-60 was 0.5%, solid-to-liquid ratio was 1:25, temperature was 60℃, and the extraction time was 30 min. The extraction yield of desertliving cistanche polysaccharides was 8.17%. It was concluded that this extraction technology was reasonable and reliable, which can be used in the development and utilization of cistanche.

ABSTRACT

Microbiologically induced deterioration (MID) causes corrosion of concrete by producing acids (including organic and inorganic acids) that degrade concrete components and thus compromise the integrity of sewer pipelines and other structures, creating significant problems worldwide. Understanding of the fundamental corrosion process and the causal agents will help us develop an appropriate strategy to minimize the costs in repairs. This review presents how microorganisms induce the deterioration of concrete, including the organisms involved and their colonization and succession on concrete, the microbial deterioration mechanism, the approaches of studying MID and safeguards against concrete biodeterioration. In addition, the uninvestigated research area of MID is also proposed.

ABSTRACT

Sulfuric acid can cause local or systemic effects after exposure by inhalation, ingestion, or topical application. Direct ingestion is the main exposure route for fatal sulfuric acid injury. Fatal accidents involving the inhalation of toxic sulfuric acid vapors are rare. Inhalation of sulfuric acid fumes causes severe irritation or corrosive damage to the upper respiratory tract. Consequently, severe congestion, edema, and inflammation of the mucous membranes of the upper respiratory passages hinder the entry of air into the lungs, possibly leading to fatalities in victims. A 35-year-old man died at his home after complaining of a severe sore throat. One day earlier, he had worked in a sulfuric acid tank in a copper-smelting plant while wearing an ordinary gas mask that offered no protection against sulfurous acid vapors. Upon autopsy, the larynx and epiglottis showed pronounced edema, congestion, and inflammation that histologically mimicked an acute bacterial suppurative inflammation, accompanied by severe pulmonary edema. A field analysis of the air inside the sulfuric acid tank revealed SO4(2-) positivity and a 40% sulfuric acid concentration.
